The Antibiotic Zone Reader quickly and accurately helps figure out how strong antibiotics are by measuring the size of the zone where bacteria can’t grow in a Petri dish.
1.0 Objective: The objective of this Standard Operating Procedure is to provide a written procedure for the operation and calibration of the Antibiotic Zone Reader.
2.0 Scope: This SOP covers the operation and calibration of the Antibiotic Zone Reader.
3.0 Responsibility: Microbiologist: Responsible for implementation of this SOP.
QA Officer/QAM: Review and Overall compliance with this SOP.
4.0 Procedure:
4.1 Operation:
4.1.1 Ensure that the instrument is visibly clean and free from dust.
4.1.2 Switch ‘ON’ the mains switch.
4.1.3 Turn the left-side drum to its maximum clockwise position, aligning the Zero Marking on the scale of the right-side drum with the Zero marking on the Vernier scale.
4.1.4 Place the Petri dish on the Aluminum plate and manually rotate it so that one end of the Zone image touches the Mark Line on the Prism.
4.1.5 Slowly turn the left-side drum anti-clockwise until the other end of the Zone image touches the Mark Line on the Prism.
4.1.6 Note the reading directly on the Right-side drum scale.
4.1.7 The scale is marked from 0 to 35 mm with each division of 0.2 mm and the Vernier scale is marked with each division of 0.02 mm.

5.0 Calibration:
5.1 Operate the instrument as per the above procedure.
5.2 Place a standard coin in the appropriate location on the antibiotic zone reader.
5.3 Measure the zone of the standard coin and compare it with both the standard coin result and the observed result.
5.4 Record the results in the calibration format.
6.0 Maintenance:
6.1 Clean the instrument with a dry cloth.
6.2 Clean the glass and lance with a dry cloth or tissue paper.
