Understand how to perform the pharmaceutical solubility test, necessary equipment, selection of solvents, classifications of solubility and reporting procedure for the QC lab.
Scope: This procedure is applicable to the solubility test.
Apparatus:
- Test tube
- Volumetric flask
Reagents:
As described in the respective specification
Method :
- Take about 1 g of sample in test tube / volumetric flask (as applicable).
- Select the solvent as per specification.
- Based on the solubility of the sample, add the required volume of the particular solvent(s) as per table-I given below.
- Shake well to dissolve ensuring that the test is carried out at a temperature between 20°C to 30°C.
- Check sample against the specification for solubility.

Table-1
| Description term | Approximate volume of solvent in milliliters per gram of solute |
| Very soluble | Less than 1 |
| Freely soluble | From 1 to 10 |
| Soluble | From 10 to 30 |
| Sparingly soluble | From 30 to 100 |
| Slightly soluble | From 100 to 1000 |
| Very slightly soluble | From 1000 to 10000 |
| Insoluble or practically insoluble | More than 10000 |
